This variant lies in the MYH2 gene (transcript NM_017534.6) at coding-DNA position 322, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 108 with lysine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C55"). This sequence change replaces glutamic acid with lysine at codon 108 of the MYH2 protein (p.Glu108Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine.
