NM_006231.4(POLE):c.5118G>T (p.Met1706Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M1706I variant (also known as c.5118G>T), located in coding exon 38 of the POLE gene, results from a G to T substitution at nucleotide position 5118. The methionine at codon 1706 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
