NM_004168.4(SDHA):c.296A>G (p.His99Arg) was classified as Uncertain significance for Pheochromocytoma/paraganglioma syndrome 5; Mitochondrial complex II deficiency, nuclear type 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine with arginine at codon 99 of the SDHA protein (p.His99Arg). The histidine residue is highly conserved and there is a small physicochemical difference between histidine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in an individual affected with jugular paraganglioma (PMID: 28384794). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
