Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.5315T>A (p.Val1772Asp), citing Ambry Variant Classification Scheme 2023: The p.V1751D variant (also known as c.5252T>A), located in coding exon 37 of the NF1 gene, results from a T to A substitution at nucleotide position 5252. The valine at codon 1751 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
