This variant lies in the BTK gene (transcript NM_000061.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1751, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Donor and acceptor splice site variants typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in BTK are known to be pathogenic (PMID: 15661032, 16862044, 19419768). Disruption of this splice site has been observed in several individuals affected with X-linked agammaglobulinemia (PMID: 16712653,9106525, 15661032). This variant is not present in population databases (ExAC no frequency). This sequence change affects an acceptor splice site in intron 17 of the BTK gene.
