NM_000264.5(PTCH1):c.1625A>G (p.Lys542Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K542R variant (also known as c.1625A>G), located in coding exon 12 of the PTCH1 gene, results from an A to G substitution at nucleotide position 1625. The lysine at codon 542 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
