Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.3331A>G (p.Met1111Val), citing Ambry Variant Classification Scheme 2023: The p.M1111V variant (also known as c.3331A>G), located in coding exon 16 of the BLM gene, results from an A to G substitution at nucleotide position 3331. The methionine at codon 1111 is replaced by valine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
