This variant lies in the CASP10 gene (transcript NM_032977.4) at coding-DNA position 199, where G is replaced by T; at the protein level this means replaces alanine at residue 67 with serine — a missense variant. Submitter rationale: This sequence change replaces alanine with serine at codon 67 of the CASP10 protein (p.Ala67Ser). The alanine residue is weakly conserved and there is a moderate physicochemical difference between alanine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CASP10-related disease.
