NM_000071.3(CBS):c.538G>A (p.Val180Met) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V180M variant (also known as c.538G>A), located in coding exon 5 of the CBS gene, results from a G to A substitution at nucleotide position 538. The valine at codon 180 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
