Uncertain significance for Charcot-Marie-Tooth Neuropathy X; Combined oxidative phosphorylation deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004208.4(AIFM1):c.1710C>A (p.Asp570Glu), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with AIFM1-related disease. This sequence change replaces aspartic acid with glutamic acid at codon 570 of the AIFM1 protein (p.Asp570Glu). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and glutamic acid. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
