Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.2654A>G (p.Asn885Ser), citing Ambry Variant Classification Scheme 2023: The p.N885S variant (also known as c.2654A>G), located in coding exon 16 of the DICER1 gene, results from an A to G substitution at nucleotide position 2654. The asparagine at codon 885 is replaced by serine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
