Likely pathogenic — the classification assigned by GeneDx to NM_173551.5(ANKS6):c.1973-3C>G, citing GeneDx Variant Classification (06012015): The c.1973-3C>G variant in the ANKS6 gene has been reported previously in a homozygous state in an individual with infantile-onset polycystic kidney disease, aortic and pulmonary stenosis, and periportal liver fibrosis (Hoff et al., 2013). This variant damages the splice acceptor site in intron 10, and is expected to cause abnormal gene splicing. The c.1973-3C>G variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). We interpret c.1973-3C>G as a likely pathogenic variant.
