NM_007315.4(STAT1):c.970T>C (p.Cys324Arg) was classified as Likely pathogenic for Autoimmune enteropathy and endocrinopathy - susceptibility to chronic infections syndrome by Illumina Laboratory Services, Illumina, citing ICSLVariantClassificationCriteria RUGD 01 April 2020: The STAT1 c.970T>C (p.Cys324Arg) variant has been reported in a heterozygous state in at least five individuals with chronic mucocutaneous candidiasis disease or multiple autoimmune diseases (Sharfe et al. 2014; Toubiana et al. 2016; Second et al. 2017; Maeshima et al. 2019). In one family, the variant was shown to be de novo in the proband and in at least two families, more than one affected individual carried the variant (Toubiana et al. 2016; Sharfe et al. 2017). Control data are unavailable for this variant which is absent from the Genome Aggregation Database. Using alanine scanning mutagenesis and GAS reporter assays, the p.Cys324Arg variant was associated with an increase in transcription in U3C cells compared to wildtype, suggesting this variant causes a gain-of-function (Kagawa et al. 2017). Based on the collective evidence, the p.Cys324Arg variant is likely pathogenic for chronic mucocutaneous candidiasis disease.
