Uncertain significance for Deafness-lymphedema-leukemia syndrome; Monocytopenia with susceptibility to infections — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032638.5(GATA2):c.803G>T (p.Gly268Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the GATA2 gene (transcript NM_032638.5) at coding-DNA position 803, where G is replaced by T; at the protein level this means replaces glycine at residue 268 with valine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with GATA2-related disease. This sequence change replaces glycine with valine at codon 268 of the GATA2 protein (p.Gly268Val). The glycine residue is moderately conserved and there is a moderate physicochemical difference between glycine and valine. This variant is not present in population databases (ExAC no frequency).
