This variant lies in the MYOM1 gene (transcript NM_003803.4) at coding-DNA position 2317, duplicating one base; at the protein level this means shifts the reading frame starting at isoleucine residue 773, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Ile773Asnfs*15) in the MYOM1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in MYOM1 cause disease.
