Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.1323G>T (p.Glu441Asp), citing Ambry Variant Classification Scheme 2023: The p.E441D variant (also known as c.1323G>T), located in coding exon 2 of the TERT gene, results from a G to T substitution at nucleotide position 1323. The glutamic acid at codon 441 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
