This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 154, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 52 with glutamine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid with glutamine at codon 52 of the MSH6 protein (p.Glu52Gln). The glutamic acid residue is weakly conserved and there is a small physicochemical difference between glutamic acid and glutamine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
