This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 500 through coding-DNA position 503, deleting 4 bases; at the protein level this means shifts the reading frame starting at threonine residue 167, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.500_503delCAAA pathogenic mutation, located in coding exon 6 of the BRCA1 gene, results from a deletion of 4 nucleotides at nucleotide positions 500 to 503, causing a translational frameshift with a predicted alternate stop codon (p.T167Sfs*66). This variant was reported in 1/60,466 breast cancer cases and in 0/53,461 controls (Dorling et al. N Engl J Med 2021 02;384:428-439). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
