NM_006231.4(POLE):c.1949C>T (p.Thr650Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T650I variant (also known as c.1949C>T), located in coding exon 18 of the POLE gene, results from a C to T substitution at nucleotide position 1949. The threonine at codon 650 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
