Pathogenic for Gastrointestinal stromal tumor; Paragangliomas 4; Pheochromocytoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003000.3(SDHB):c.148_151dup (p.Lys51fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SDHB gene (transcript NM_003000.3) at coding-DNA position 148 through coding-DNA position 151, duplicating 4 bases; at the protein level this means shifts the reading frame starting at lysine residue 51, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Lys51Argfs*13) in the SDHB gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). This variant has been observed in several individuals affected with paraganglioma (PMID: 23154831). This variant is also known as c.151_154insGACA in the literature. Loss-of-function variants in SDHB are known to be pathogenic (PMID: 19454582, 19802898). For these reasons, this variant has been classified as Pathogenic.
