Uncertain significance for Nephronophthisis 16 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_173551.5(ANKS6):c.664C>G (p.Arg222Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 222 of the ANKS6 protein (p.Arg222Gly). This variant is present in population databases (rs41283630, gnomAD 0.1%). This variant has not been reported in the literature in individuals affected with ANKS6-related conditions. ClinVar contains an entry for this variant (Variation ID: 641730). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
