Uncertain significance for Immunodeficiency 26 with or without neurologic abnormalities — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006904.7(PRKDC):c.9811G>A (p.Asp3271Asn), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PRKDC gene (transcript NM_006904.7) at coding-DNA position 9811, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 3271 with asparagine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid with asparagine at codon 3271 of the PRKDC protein (p.Asp3271Asn). The aspartic acid residue is modrately conserved and there is a small physicochemical difference between aspartic acid and asparagine. This variant is present in population databases (rs780788138, ExAC 0.006%). This variant has not been reported in the literature in individuals with PRKDC-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
