Pathogenic for Griscelli syndrome type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000015.10:g.(?_55223869)_(55234954_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in RAB27A are known to be pathogenic (PMID: 10835631, 23160464). A similar deletion of exons 2-5 has been observed in an individual affected with¬†primary hemophagocytic lymphohistiocytosis¬†(PMID:¬†16278825). This variant is a gross deletion of the genomic region encompassing exons 2-5 of the RAB27A gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 5 of the RAB27A gene. This is expected to result in an absent or disrupted protein product.
