NM_000548.5(TSC2):c.4849+6_4849+27del was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Sema4, Sema4, citing Sema4 Curation Guidelines. This variant lies in the TSC2 gene (transcript NM_000548.5) at 6 bases into the intron immediately after coding-DNA position 4849 through 27 bases into the intron immediately after coding-DNA position 4849, deleting this region. Submitter rationale: The TSC2 c.4849+6_4849+27del variant has not been reported in the literature to our knowledge. It was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID 641528). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing, but this prediction has not been confirmed by published transcriptional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
