Uncertain significance for Pancreatic adenocarcinoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001166108.2(PALLD):c.3090T>G (p.Ile1030Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine with methionine at codon 526 of the PALLD protein (p.Ile526Met). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and methionine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with PALLD-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
