Uncertain significance for Charcot-Marie-Tooth disease type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002047.4(GARS1):c.2008G>C (p.Val670Leu), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs376585160, ExAC 0.01%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with GARS-related disease. This sequence change replaces valine with leucine at codon 670 of the GARS protein (p.Val670Leu). The valine residue is weakly conserved and there is a small physicochemical difference between valine and leucine.
