Uncertain significance — the classification assigned by Ambry Genetics to NM_003803.4(MYOM1):c.74C>A (p.Thr25Asn), citing Ambry Variant Classification Scheme 2023: The p.T25N variant (also known as c.74C>A), located in coding exon 1 of the MYOM1 gene, results from a C to A substitution at nucleotide position 74. The threonine at codon 25 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
