Uncertain significance for Developmental and epileptic encephalopathy, 1; Autosomal recessive spinocerebellar ataxia 12 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016373.4(WWOX):c.605T>G (p.Val202Gly), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the WWOX gene (transcript NM_016373.4) at coding-DNA position 605, where T is replaced by G; at the protein level this means replaces valine at residue 202 with glycine — a missense variant. Submitter rationale: This sequence change replaces valine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 202 of the WWOX protein (p.Val202Gly). This variant is present in population databases (rs112636835, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with WWOX-related conditions. ClinVar contains an entry for this variant (Variation ID: 641170). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
