Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000393.5(COL5A2):c.553G>A (p.Asp185Asn), citing Ambry Variant Classification Scheme 2023: The p.D185N variant (also known as c.553G>A), located in coding exon 7 of the COL5A2 gene, results from a G to A substitution at nucleotide position 553. The aspartic acid at codon 185 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
