NM_015272.5(RPGRIP1L):c.946C>T (p.Gln316Ter) was classified as Pathogenic for Joubert syndrome; Meckel-Gruber syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in RPGRIP1L are known to be pathogenic (PMID: 17558409). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has not been reported in the literature in individuals with RPGRIP1L-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Gln316*) in the RPGRIP1L gene. It is expected to result in an absent or disrupted protein product.
