NM_001005242.3(PKP2):c.1379-1976G>A was classified as Uncertain Significance for Arrhythmogenic right ventricular cardiomyopathy by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This variant causes a G>A nucleotide substitution at the +1 position of intron 6 of the PKP2 gene. Splice prediction tools suggest that this variant may disrupt splicing. This variant is predicted to cause skipping of exon 6 of the PKP2 transcript NM_004572.3. However, this exon is alternatively spliced and is absent in the predominant isoform expressed in the heart (NM_001005242, PMID: 21378009). To our knowledge, functional studies have not been performed for this variant. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
