Uncertain significance for Disseminated atypical mycobacterial infection — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000416.3(IFNGR1):c.866C>G (p.Ser289Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with cysteine at codon 289 of the IFNGR1 protein (p.Ser289Cys). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and cysteine. This variant is present in population databases (rs775472233, ExAC 0.01%). This variant has not been reported in the literature in individuals with IFNGR1-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
