NM_001370259.2(MEN1):c.1621A>C (p.Thr541Pro) was classified as Uncertain significance for Multiple endocrine neoplasia, type 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with MEN1-related disease. This sequence change replaces alanine with proline at codon 541 of the MEN1 protein (p.Ala541Pro). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and proline. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Not available"; Align-GVGD: "Class C0". The proline amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function.
