NM_005359.6(SMAD4):c.3G>C (p.Met1Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015: This variant results in the loss of the translation initiator methionine at codon 1 of the SMAD4 protein. However, codons 4 and 24 encode for methionine that may serve as alternative translation start sites. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with SMAD4-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
