This variant lies in the NBN gene (transcript NM_002485.5) at coding-DNA position 980, where G is replaced by T; at the protein level this means replaces glycine at residue 327 with valine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with valine at codon 327 of the NBN protein (p.Gly327Val). The glycine residue is weakly conserved and there is a moderate physicochemical difference between glycine and valine. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
