Uncertain significance for Bailey-Bloch congenital myopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_145064.3(STAC3):c.997-1G>C, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the STAC3 gene (transcript NM_145064.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 997, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change falls in intron 11 of the STAC3 gene. It does not directly change the encoded amino acid sequence of the STAC3 protein. It affects a nucleotide within the consensus splice site of the intron. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with STAC3-related conditions. Nucleotide substitutions within the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
