Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002528.7(NTHL1):c.743C>G (p.Thr248Ser), citing Ambry Variant Classification Scheme 2023: The p.T256S variant (also known as c.767C>G), located in coding exon 5 of the NTHL1 gene, results from a C to G substitution at nucleotide position 767. The threonine at codon 256 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
