NC_000003.12:g.(?_167684288)_(167720177_?)del was classified as Pathogenic for Cerebral cavernous malformations 3 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the PDCD10 gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. Similar deletions of PDCD10 (also known as the CCM3 gene) have been observed in individuals affected with cavernous malformations (PMID: 18060436, 20623299) and one larger deletion including neighboring genes occurred de novo (PMID: 15543491). Loss-of-function variants in PDCD10 are known to be pathogenic (PMID: 15543491, 18300272, 23801932). For these reasons, this variant has been classified as Pathogenic.
