NM_001457.4(FLNB):c.482T>G (p.Phe161Cys) was classified as Likely pathogenic by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces phenylalanine, which is neutral and non-polar, with cysteine, which is neutral and slightly polar, at codon 161 of the FLNB protein (p.Phe161Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Larsen syndrome (PMID: 14991055). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 6398). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on FLNB protein function. Experimental studies have shown that this missense change affects FLNB function (PMID: 22190451). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
