NM_022356.4(P3H1):c.466A>G (p.Ile156Val) was classified as Uncertain significance for Osteogenesis imperfecta type 8 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine with valine at codon 156 of the P3H1 protein (p.Ile156Val). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and valine. This variant is present in population databases (rs751518628, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with P3H1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
