This variant lies in the PREPL gene (transcript NM_001171613.2) at coding-DNA position 760, where A is replaced by G; at the protein level this means replaces methionine at residue 254 with valine — a missense variant. Submitter rationale: This sequence change replaces methionine with valine at codon 343 of the PREPL protein (p.Met343Val). The methionine residue is moderately conserved and there is a small physicochemical difference between methionine and valine. This variant is present in population databases (rs771442588, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with PREPL-related conditions.
