Pathogenic for Charcot-Marie-Tooth disease type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024577.4(SH3TC2):c.3321C>A (p.Tyr1107Ter), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 637416). This premature translational stop signal has been observed in individuals with Charcot-Marie-Tooth disease (PMID: 25429913, 31634715). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Tyr1107*) in the SH3TC2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SH3TC2 are known to be pathogenic (PMID: 20220177, 27068304).
