NM_000166.6(GJB1):c.499A>G (p.Lys167Glu) was classified as Uncertain significance for Charcot-Marie-Tooth disease X-linked dominant 1 by 3billion, citing ACMG Guidelines, 2015. This variant lies in the GJB1 gene (transcript NM_000166.6) at coding-DNA position 499, where A is replaced by G; at the protein level this means replaces lysine at residue 167 with glutamic acid — a missense variant. Submitter rationale: The variant is not observed in the gnomAD v4.1.0 dataset. Predicted Consequence/Location: Missense variant In silico tool predictions suggest damaging effect of the variant on gene or gene product [REVEL: 0.90 (>=0.6, sensitivity 0.68 and specificity 0.92); 3Cnet: 0.87 (> 0.75, sensitivity 0.96 and precision 0.92)]. The same nucleotide change resulting in the same amino acid change has been previously reported to be associated with GJB1-related disorder (PMID: 14960772). However, the evidence of pathogenicity is insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
