NM_018972.4(GDAP1):c.840del (p.Tyr279_Tyr280insTer) was classified as Pathogenic for Charcot-Marie-Tooth disease; Charcot-Marie-Tooth disease type 4A by Sadaf Naz Human Genetics Laboratory, University of the Punjab, citing ACMG Guidelines, 2015. This variant lies in the GDAP1 gene (transcript NM_018972.4) at coding-DNA position 840, deleting one base. Submitter rationale: The variant Tyr280Ter causes Charcot-Marie-Tooth disease (CMT). Patients exhibit gait disturbances, hand and feet contractures, valgus wrist and ankles. Patients had atrophy in distal limb muscles. The variant is absent in the gnomAD and 600 chromosomes of the ethnically matched control population. The variant had also been reported previously in another individual having delayed developmental milestones, muscle atrophy of hands and distal legs and a bilateral foot drop. In summary, we classify the variant Tyr280Ter as pathogenic based on our segregation analysis and absence of the variant in public databases and ethnically matched control population.
