This variant lies in the MYBPC3 gene (transcript NM_000256.3) at the canonical splice donor site of the intron immediately after coding-DNA position 1457, where T is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.1457+2T>G intronic variant results from a T to G substitution two nucleotides after coding exon 16 in the MYBPC3 gene. This alteration has been reported in a hypertrophy cardiomyopathy (HCM) genetic testing cohort (Hathaway J et al. BMC Cardiovasc Disord, 2021 Mar;21:126). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site. In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay.
