Uncertain significance for Surfactant metabolism dysfunction, pulmonary, 4 — the classification assigned by Johns Hopkins Genomics, Johns Hopkins University to NM_172245.4(CSF2RA):c.281T>C (p.Phe94Ser), citing ACMG Guidelines, 2015: This CSF2RA variant (rs774500472) is rare (<0.1%) in large population datasets (gnomAD: 4/282562 total alleles; 0.001%; no homozygotes). CSF2RA c.281T>C has not been reported in ClinVar nor the literature, to our knowledge. Two bioinformatic tools queried predict that this substitution would possibly be damaging, however the phenylalanine residue at this position is not highly evolutionarily conserved across the species assessed. Bioinformatic analysis predicts that this variant would not affect normal exon 6 splicing, although this has not been confirmed experimentally to our knowledge. The clinical significance of c.281T>C is uncertain at this time.
