NM_001089.3(ABCA3):c.2411A>G (p.His804Arg) was classified as Uncertain significance for Interstitial lung disease due to ABCA3 deficiency by Johns Hopkins Genomics, Johns Hopkins University, citing ACMG Guidelines, 2015: This ABCA3 variant (rs747443485) is rare in large population datasets (gnomAD: 1/249380 total alleles; 0.0004010%; no homozygotes). This variant has not been reported in ClinVar nor the literature, to our knowledge. Of two bioinformatics tools queried, one predicts that this substitution would be benign, while the second predicts that it would be deleterious. Additionally, the histidine residue at position 804 is evolutionarily conserved across most species assessed. Bioinformatic analysis predicts that this variant would not affect normal exon 18 splicing, although this has not been confirmed experimentally to our knowledge. The clinical significance of c.2411A>G is uncertain at this time.
