NM_000159.4(GCDH):c.1018C>T (p.Leu340Phe) was classified as Uncertain significance for Glutaric aciduria, type 1 by 3billion, citing ACMG Guidelines, 2015. This variant lies in the GCDH gene (transcript NM_000159.4) at coding-DNA position 1018, where C is replaced by T; at the protein level this means replaces leucine at residue 340 with phenylalanine — a missense variant. Submitter rationale: The variant is observed at an extremely low frequency in the gnomAD v4.1.0 dataset (total allele frequency: <0.001%). Predicted Consequence/Location: Missense variant In silico tool predictions suggest damaging effect of the variant on gene or gene product [REVEL: 0.81 (>=0.6, sensitivity 0.68 and specificity 0.92)]. The same nucleotide change resulting in the same amino acid change has been previously reported to be associated with GCDH related disorder (PMID: 32777384). However, the evidence of pathogenicity is insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
