Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4.2e-05 in 1613020 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in GBA1 causing Gaucher Disease (4.2e-05 vs 0.005), allowing no conclusion about variant significance. c.1495G>C has been reported in the literature in several individuals affected with Gaucher-related Parkinsons' disease without clear clinical information or with a complex genotype in 1 individual with Gaucher disease without clinical information (McNeill_2012, Machaczka_2018, Petrucci_2020, Kars_2021). These report(s) do not provide unequivocal conclusions about association of the variant with Gaucher Disease.
