This variant lies in the PLOD1 gene (transcript NM_000302.4) at coding-DNA position 613, where C is replaced by T; at the protein level this means replaces arginine at residue 205 with cysteine — a missense variant. Submitter rationale: Variant summary: PLOD1 c.613C>T (p.Arg205Cys) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 3.6e-05 in 251414 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.613C>T in individuals affected with Ehlers-Danlos syndrome, kyphoscoliotic type 1 and no experimental evidence demonstrating its impact on protein function have been reported.
